Prognostic factors of non-muscle invasive bladder cancer: a study based on next-generation sequencing

Abstract Objective To investigate the genetic prognostic factors for recurrence of non-muscle invasive bladder cancer. Materials and methods The patients underwent transurethral resection tumor received bacillus Calmette–Guérin (BCG) or epirubicin. Next-generation sequencing was performed alterations genes, pathways, mutation burden were recorded. Associations between these clinicopathological variants estimated, factor identified. Results A total 58 cases included in our study, 46 treatment with BCG. FGFR3 most frequently altered gene (48%), more commonly detected intermediate-risk patients. Univariate Cox analysis demonstrated that 10 genes significantly correlated BCG failure, while NEB, FGFR1 SDHC independent predictors. Besides, epigenetic-related pathway mutations negatively (hazard ratio: 0.198, P = 0.023). DNA damage response repair positively burden, TP53 frequent among significant high burden. Conclusion instillation reduced rate compared epirubicin this population. Potential biomarkers therapeutic targets found help next-generation sequencing; correlations DDR also demonstrated.
